Prep Column Packing: SEM Evidence for Mechanical Durability

Examine particle morphology after repeated packing stress before committing to downstream selectivity and recovery testing.

Packing a dynamic axial compression (DAC) column for prep liquid chromatography (LC) is a half-day commitment that assumes the bed will survive multiple separation cycles. With conventional silica, that operational lifespan is often cut short. Fines from fractured particles work into the frits and backpressure climbs, sacrificing run capacity to a physical problem that has nothing to do with the chemistry.

This application note documents the packing properties of YMC-Triart Prep—a hybrid-silica phase tested against conventional silica under repeated DAC packing and against itself under hydraulic compression at scale-up pressures. Scanning electron microscopy (SEM) at named cycle counts reveals where conventional silica shatters into fines and where the hybrid-silica phase keeps its bead structure intact.
